Optimal secretion of the recombinant protein can be obtained by a desirable signal peptide .In this research, we used Bacillus. sp endoxylanase signal peptide in order to guide the human growth hormone (HGH) to the periplasmic space as an effective approach for production of an active and non-immunogenic form of rHGH without formyl methionine at the N-terminus. The signal sequence was fused to the growth hormone gene and cloned in pET 28a+ vector under the T7 promoter and transformed to Ecoli، Rosetta gami and BL21 (DE3) strains. The bacterial cells in log phase indused by 0.25 mM of IPTG. Osmotic shock used to releas periplasmic components followed by SDS-PAGE and eventually confirmed by western blot. The results showed a high portion of rHGH transferred to the periplasmic space that means xylA signal peptide successfully led to the protein to periplasm. Among the tested constructs, pET28a+xyla-rHGH in E. coli Rosetta gami had 12.49 % of rHGH band depending on Imag J software analysis. The aim of this study was to compare the effects of silver nanoparticles and alcoholic extract of Amaranthus retroflexus on some pathogenic bacteria. The alcoholic extract of this plant was prepared and the effect of concentrations of 50 to 400 mg/ml of this extract and concentrations of 10 to 80 mg/ml of silver nanoparticles by well diffusion method on S. aureus, B. cereus, E coli and P aeruginosa were evaluated. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined on bacteria by dilution method. The results showed that the inhibition effect of Amaranthus retroflexus on gram-positive bacteria is more than gram negative bacteria while the inhibitory effect of silver nanoparticles on gram-negative bacteria is more than gram-positive bacteria. The results of this study showed that the alcoholic extract of Amaranthus retroflexus had an antibacterial properties that can be considered as an adjunct to further studies for in vivo conditions and replacement with common chemical agents in the treatment of infections.Health Biotechnology and BiopharmaHealth Biotechnology and Biopharma (HBB)2538-44142320181201Investigation of slow freezing of human testicular tissue on proliferation, colonization and viability of human spermatogonial stem cells243513347710.22034/HBB.2018.24ENJournal Article20210713In this study, the effects of freezing of testicular tissue by slow freezing on the proliferation rate, colonization and viability of spermatogonial stem cells (SSCs) were evaluated. The testicular tissue of the freezing group was freezed by slow-freezing method, and the fresh group remained intact. The results showed that the cell proliferation rates of fresh group [1.48 ± 0.02] and [1.77 ± 0.02] compared to the frozen-thawed group [1.39 ± 0.02] and [1.64 ± 1.64] in the 7th and 14th days, were significantly higher. There was significant difference between the number of colonies in the 7th and 14th days. In the first day after cell isolation, cell viability of the fresh group [95 %] was significantly higher than the frozen-thawed group [87.7 %]. Also, in the 7th and 14th day, cells viability fresh group were significantly more than frozen-thawed group. From 1989 to 2013 in a single blind randomized clinical trial conducted in Shiraz central hospital, 700 cases (460 females and 240 males) with the diagnosis of moderate to severe pain intensity migraine were enrolled. The cases (n=350) were put on vitamin C (ascorbic acid) 25 mg in a capsule mixed in a cup of tea (100 ml) containing about 20 mg caffeine (Migracorb), to drink three times per day with sugar. The controls (n=350) drank three times per day with sugar. 22 % of the female cases and 7.5 % of the male cases had abortive response to Migracorb from 10 min to 3 h. 53 % of the female cases and 39 % of the male cases were cured by taking Migracorb after 6 months. Only 0.5 % of the female control group cases were cured after 6 months.<br />The combination of vitamin C as antioxidant and caffeine as anti-inflammation cured a significant number of patients with migraine headache.Health Biotechnology and BiopharmaHealth Biotechnology and Biopharma (HBB)2538-44142320181201Assessing the frequency of t (12; 21) (p13; q22) in patients with acute lymphoblastic leukemia in northeastern of Iran486213347910.22034/HBB.2018.26ENJournal Article20210713Studying the results of the prevalence of a particular genetic translocation of acute lymphoblastic leukemia (ALL) could predict the incidence rate of overall survival (OS) and event-free survival (EFS). 283 patients from northeastern of Iran with new diagnosis of ALL by bone marrow aspiration or peripheral blood sample were enrolled in the study. The expression of TEL-AML1 fusion were studied by TaqMan probe qualitative real time PCR compared with ABL reference gene. Then, the five-year survival rates were surveyed in the region of Khorasan Razavi and were analyzed by kaplan-meier test. Prevalence of t (12, 21) in patients with ALL was 14.4 %, survival rate was 75 %, and the average five-year survival rate for patients was 4.77 ± 39.76 months by the log-rank test. Prevalence of t (12, 21) in patients with ALL in the north east of Iran was lower than the world average.Health Biotechnology and BiopharmaHealth Biotechnology and Biopharma (HBB)2538-44142320181201Vitamin E reduces blood urea concentrations level after formaldehyde exposure in rat636713348010.22034/HBB.2018.27ENJournal Article20210713In this study, effect of vitamin E on kidney after exposure to formaldehyde was evaluated. 24 adult male wistar rats divided randomly in three groups: control, formaldehyde (E1), vitamin E and formaldehyde (E2). After two weeks rats were sacrificed, and 5 ml of blood was taken from their heart. Blood urea nitrogen (BUN) and creatinine concentrations were measured. BUN concentration was significantly increased in E1 (P=0.03) and E2 groups (P=0.02) compare with control group. There were no significant differences in creatinine levels between groups (p=0.11). The results showed that vitamin E probably protect kidney against formaldehyde alterations in rat. It may be possible to use vitamin E when increasing blood urea concentrations to restore urea plasma balance and prevent kidney damage.
